    Further, I also gained wet lab hands-on training by attending summer research camps and conferences that helped to foster curiosity and scientific undertakings, For instance, I took a wet lab training summer internship at IISC Bangalore, affiliated lab Protein design under Dr Deepti Saini through which I grain knowledge of Primer designing using BLAST, molecular cloning, restriction digestion in C41 Bacterial plasmids and Identification pET28a, pET24a expression plasmids and, I completed a two-week wet lab training workshop at the National Brain Research Centre in Haryana, where I earned certification in Laboratory Animal Science (CCLAS). I was trained in handling rodents, B form filling, animal welfare guidelines, 3Rs in research and conducting behavioural studies, including the administration of saline through renal, oral gavage, intraperitoneal, intramuscular, and intravenous injections. I also learned to induce Parkinson’s disease models via stereotactic surgery using 6-OHDA. Also, I gave an oral presentation titled, “Investigation of the effect of semaglutide in MPTP-induced Parkinson’s disease in mice via oral gavage” The study of neuroinflammation, along with cell signalling processes in molecular pathways, drives my interest in rodent-based translational research. I developed not only technical skills but also a strong appreciation for collaboration.

    During my Master’s thesis, I investigated the gut microbiota of Tenebrio molitor, focusing on bacteria capable of degrading polystyrene. This work enhanced my laboratory skills and deepened my understanding of plastivore microbiology, while also sparking my interest in the gut-brain axis of interdisciplinary research. My curiosity brought me to the Centre for Neuroscience in Kochi, Kerala, in 2023, where I worked as a Project Assistant under Dr Baby Chakrapani P.S. At the Centre, I used a C57BL/6 mouse model to investigate how colitis affects the connection between the brain and gut. I researched how the gut microbiota and the brain interact bidirectionally, and faecal microbiota transplantation can mitigate IBD conditions. Also, research on how bile acids trigger cytotoxic stress and signalling pathways. I worked on several molecular biology methods, like Western blotting, SDS-PAGE, extracting RNA with TRIzol, RT-PCR, qPCR, FACS, immunohistochemistry, gel electrophoresis, and MTT assays.

    As I investigated colitis-induced ER stress, I have learned more about the UPR, oxidative stress, mitochondrial dysfunction, and neuroinflammation, and how they are involved in the function of the neuroimmune system. This experience has improved my research abilities, especially with molecular techniques, data analysis (T-test ), and projects that involve generating and testing a hypothesis.

    Most recently, I conducted research under Dr. Mustak on the neuroprotective potential of phytochemicals in an epileptic mutant Drosophila model. The project focused on identifying compounds such as quercetin to reduce inflammation and programmed cell death, particularly by suppressing NF-κB activity and reducing reactive oxygen species (ROS). Using climbing assays and molecular techniques, I investigate how antioxidant-rich phytochemicals exhibit anti-inflammatory effects and their potential as treatments for seizures. During this time, I also followed up on a related project previously initiated by PhD alumni: “Ionising Radiation-Induced ER Stress Response in Astrocytes and Neuronal Cell Neuroprotection by Phytochemicals (in vivo and in vitro).” This study examined how ionising radiation (IR) activates the unfolded protein response (UPR) via BIP and CHOP expression in astrocytes, leading to cellular damage and cognitive decline. These findings suggest that targeting the UPR cascade may mitigate radiation-induced neurodegeneration. Through this work, I have developed a deep appreciation for the attention to detail, discipline, and perseverance required in a research setting.

    Through my training, I have gained substantial experience in cellular and molecular biology, and I am confident that I am a strong candidate for this position, as I have adequate knowledge of rodent behavioural experiments, molecular techniques, and translational animal modelling. Further, I am eager to refine my knowledge under your mentorship. D-NOME biosciences, with its renowned translational research program, research facility, and collaborative research environment, offers the ideal setting to grow as an independent scientist.     Please accept my application for the Scientist- Molecular Biology at D-NOME biosciences. I moved to Hyderabad in Feb 2025 from Texas US where I was employed as a post-doctoral research associate with Texas A&M Institute for Advancing Health Through Agriculture. My area of expertise in the institute was focused on improving the nutritional qualities of cereal grains deploying genomics and molecular biology tools under the responsive agriculture domain.
    I earned my Ph.D. in 2020 from **BRIC- National Agri-Food Biomanufacturing Institute (BRIC-NABI)** and the Department of Biotechnology, Panjab University, Chandigarh, India. My Ph.D. thesis entitled “**Molecular and genetic basis of amylose/resistant starch variation in bread wheat (_Triticum aestivum_ L)**”. During the course of study I developed EMS mutagenised population (approx. 1200) of bread wheat with wide amylose and resistant starch variation. **This was the first report in wheat suggesting such extreme variations for the trait and the findings were published in BMC Plant Biology, 2016. **Further molecular and genetic studies for the trait were conducted to identify the factors controlling the trait. Various mapping (RIL and backcross) populations for the high amylose trait were developed in same and different genetic backgrounds. Preliminary studies to identify QTLs were performed in F2:3 segregating populations using GBS technology. The QTL-gene identified was sequenced to validate the novel mutations, to further analyze the effect of mutation in protein function the gene was cloned in bacterial system and protein expression was measured. Further to assess the substrate binding activity of the mutant enzyme in-vitro enzyme activity were performed and measured through column chromatography using AKTA PURE system. The findings from the work were published in Euphytica (Mishra et al., 2021 and Sharma et al., 2022).** From my doctoral work I identified two low glycemic wheat lines which are currently at technology readiness level 8 (TRL8) for varietal release. **
    In the past five years of my post-doctoral research, I worked on whole genome sequence data of Indian wheat ‘C 306’. The sequence data was _de novo_ assembled and analyzed to identify novel genomic variants associated with chapatti quality characteristics of the variety when compared to the published reference genomes of wheat (Chinese Spring Refseq v1.1 and 10+ wheat genomes). **This was the first high quality assembled genome of any Indian wheat variety**; the key findings from the above work were published in **‘Genomics’ 2021**. Simultaneously, I handled various other genome sequence data from Indian wheat genotypes for improved micronutrient traits. During my last post-doctoral experience at Texas A&M I have been constantly handling larger set of genomics data and continuously deploying molecular biology tools for gene validation using bacterial system.
     
    My past research exposure in academic institutions, I have gained knowledge of various biotechnological, molecular biology tools and bioinformatics to drive continuous flow of research work to achieve the objective of the project. I have thrived in this competitive research environment because I understand the art of integrating plant biology, microbiology and molecular biology. In summary I believe the blend of my Ph.D. and Postdoc expertise on working in genomics, molecular biology and bioinformatics is a complete package that provides analytical knowledge, critical thinking and research experience that helps solve the biological questions for various ongoing projects of the organization.     I am writing to express my keen interest in applying for the post of Scientist-Molecular Biology.
    I am working as Project Scientist at International Center for Genetic Engineering and Biotechnology (ICGEB)- New Delhi, currently working on a project titled entitled “Targeting drug resistance in tuberculosis and malaria”.
    Here, my role is to develop and characterize novel therapeutic scFvs against targeting _Mycobacterium tuberculosis (Mtb)_ surface protein using phage-displayed scFv libraries.
    I have successfully screened Tomlinson I, J and HuScL-4 phage-displayed human scFv libraries against _Mtb_ surface protein and their epitope to develop potential _Mtb_ neutralizing scFvs followed by cloning and purifying recombinant proteins at a milligram scale, and evaluated their binding affinity using ELISA, Biolayer Interferometry (BLI) and Flow cytometry. Further investigate the uptake reduction of selected scFvs in THP-1 monocyte-derived macrophages. Subsequently, evaluate the efficacy of selected scFvs in animal model (mice) and engineer them into alternative formats, including scFv-Fc fusion proteins and full-length monoclonal antibodies (mAbs), for further functional characterization.
    I completed my M.Tech Biotechnology specialization in Genetic Engineering from Sharda University, Greater Noida in June 2023. During my Master’s training period, I have done my dissertation project in Genomics and Integrative Biology (CSIR–IGIB) project entitled “Construction of CMPK2 Deletion Mutants and Validation of its Expression in HEK293T cells” where my responsibilities helped me to gain experience in Molecular biology and Cell and Tissue culture techniques. During my Bachelor’s training period, I took up my B.tech. final year project in the molecular biology and genomics Lab at the International Centre for Genetic Engineering, New Delhi under the supervision of Dr. Dhiraj Verma. I completed my thesis entitled “High throughput Cloning of Human and _Mtb_ gene”. This project has helped me gain research experience in Basic molecular and microbiological techniques. I must tickle my mind to solve and troubleshoot problems and analyze the results at this level. These works helped me have a deep understanding and knowledge of genomics and molecular Biology.
    I have experience of working in a startup company Biologia Research India Pvt. Ltd. (2019-2022), As the works were multidisciplinary and based on the types of samples received. So had to proceed as well as design the experiments accordingly. As a result, I have inculcated good expertise in the molecular Biology techniques (Cloning, DNA Barcoding, DNA isolation, RNA isolation, bacterial Transformation, plasmid Isolation, PCR, qRT-PCR, small scale protein expression, SDS PAGE, and western blotting).
