    Cover Letter:

I earned my [hidden] in Biotechnology in February 2021, focusing on drug discovery. My doctoral research centered on targeting β-catenin, a critical driver in 80% of sporadic colorectal cancers, through sesamin and its analogs. By leveraging rigid docking and molecular dynamics (MD) simulations, I identified potent inhibitors, which were subsequently validated through cell-based assays, flow cytometry, and Western blotting in SW480 and HT29 cell lines. This interdisciplinary approach reinforced my ability to bridge computational predictions with experimental validation.

Building upon this foundation, my postdoctoral research expanded my expertise in virtual screening using Schrödinger software and database mining through DrugBank, NCI natural database, and IBS natural compound database. Additionally, I developed doxorubicin-resistant MDA-MB-231 cell lines and conducted cytotoxicity assays to evaluate novel compounds against both standard and drug-resistant cancer cells.

My college thesis project was titled- ‘Vaccine Design Against Ebola Virus Glycoprotein Using In-silico Approaches’ in which the primary goal was to strategize a potential vaccine design against Ebola virus variants using in-silico approaches. We identified a glycoprotein sequence which was similar in two Ebola variants and its antigenicity was checked using VaxiJen [hidden] which proved that the target sequence was antigenic in nature. Physicochemical and Structural analysis was performed and it was concluded that the selected target protein can be used as a best possible way to develop into a vaccine and for further research. My dissertation project at BHABHA ATOMIC RESEARCH CENTRE was- ‘To Investigate The Effect Of Redox Modifications On Functions Of [hidden] Cells’. I had hands-on training in various Animal tissue culture techniques like passaging, subculturing, thawing, freezing and cryopreservation, Maintenance and have used various high-tech instruments like IncuCyte, Flow cytometer, GelDoc, HTS analyzer, Microplate reader, Lyophilizer and BSL and animal hood. I have performed various phagocytosis assays, efferocytosis assays, PI staining, CFSE staining, DCFDA staining, NO assay, Clonogenic assay, MTT assay, ELISA, AGE, PAGE, RT-PCR, Cell cycle analysis, Bradford assay, Imaging. I am well-versed in using different cell-lines and their handling and storing them by making cryo vials. Apart from this I also had training in animal handling and mice dissection of spleen, intestine and bone marrow of BALB/c and Swiss mice. My project included 2 antioxidants (Chlorophyllin and Baicalein) and 2 prooxidants (Plumbagin and Mitocurcumin) and their modulations on the different functions of RAW cells [hidden] macrophages and how this could be applied further if there is a malfunctioning of these cells. I had my internship training in Apollo Diagnostics and had hands on training in RT-PCR techniques for covid-19 and Manual and automated RNA isolation techniques and data entry operations. I am currently working as a Project Assistant on an ICMR funded project- “Immunometabolomics in the quest for comprehending protection and pathogenesis in tuberculosis”. My work involves understanding metabolite profiles from patient immune cells, comparing them with healthy individuals and those with latent TB. In this role, I have gained hands-on experience in patient blood collection, Density gradient centrifugation, PBMC’s isolation and culture maintenance, Metabolite extraction, analyzing mass spectrum results, Isolation of T-cells by Magnetic associated cell separation, T-cell expansion, Intracellular staining for Flow cytometry, as well as REMA assay, Heat induced Hemolysis and Docking. I am a quick learner who thrives in hands-on laboratory environments and am eager to expand my knowledge in microfluidics and 3D cancer modelling. My strong analytical skills, attention to detail, and ability to adapt to interdisciplinary research make me a perfect fit for this role.

    Cover Letter:

Sure! Here is the redacted text:

    Dear Sir,
I am M. Angel Nivya, a graduate student of Prof. Rajakumara Eerappa from the Indian Institute of Technology (IIT) Hyderabad, India. I had a working experience of 3 years in the Centre for cellular and molecular biology (CCMB) under the guidance of Dr. Puran Singh Sijwali and worked as a research associate in the Institute of Genomics and Integrative Biology (IGIB) under the guidance of Dr. Archana Singh for one year. I was working as Project scientist II in the Translational Health Science and Technological Institute (THSTI) under the guidance of Dr Milan Surjit.
Research experience from [hidden] expertise (in research) belongs to the cloning and purification of proteins and their biophysical and computational approaches to investigate the molecular interactions of proteins to histone peptides. Most of my [hidden] research work focused on the UHRF1 (Ubiquitin-like, containing PHD and RING finger domain). We have demonstrated that the binding affinities of UHRF1 TTD exhibit higher binding affinity towardsH3K9me2 compared to H3K9me1/3through isothermal titration calorimetry (ITC). We have employed a computational tool, Molecular Dynamic (MD) simulation, to characterize the methyl status-specific binding of H3R2K9me by the TTD-PHD, and to analyze the effects of H3K4 di-methylation on H3R2K9me recognition. We have validated the MD simulation data through ITC binding study. These studies establish that the UHRF1 TTD-PHD exhibits lower binding free energy (ΔGbind) towards H3R2K9me2 compared to H3R2K9me3. We also showed that the SRA domain of UHRF1 binds to all 5mC oxidation derivatives.
Research Associate: In CCMB, I was working on biochemical and biophysical characterization of DNA Damage inducible (DDi1) protein from Plasmodium falciparum. Ddi1 is a retroviral aspartyl protease and is involved in the repair of DNA-protein crosslink. Ddi1 also acts as a proteasome shuttle factor. We investigated the Plasmodium DDI1 to identify its roles during parasite development and potential as a therapeutic target. We have also knocked in pfDDi 1 into Saccharomyces cerevisiae to investigate the growth effect.
In IGIB, I was working on isolation of Keratinocytes, Melanocytes and Fibroblast cells from foreskin patient samples and Cloning, Expression and purification of Transglutaminase 1(TGM1) protein from [hidden]
In THSTI, I was involved in the Development of a recombinant vaccine against Hepatitis E Virus and their immunological characterization.
Skills in Brief: I have also trained in molecular biology, biochemistry, and structural biology related techniques and also experienced in mammalian cell culture, Saccharomyces cerevisiae and Pichia pastoris. I am very well experienced in protein biochemistry, biochemical and biophysical characterization of proteins and epigenetics related concepts and experiments.
Research Interest: I am interested in DNA- protein, RNA- Protein and peptide-protein interactions, protein biochemistry, Structural biology and Epigenetics. My main goal is to setup independent research lab in good academic institute.
I believe that based on my research experience and skill set gained, I would perform to the best of ability for the given project. I am a highly motivated, innovative individual who works well independently as well as collaboratively on teams of scientists from various backgrounds. I also had experience in mentoring and training both undergraduate and postgraduate students with little prior research skills.
    Thank you.
    Sincerely,
    Dr. M. Angel Nivya.
