    My college thesis project was titled- ‘Vaccine Design Against Ebola Virus Glycoprotein Using In-silico Approaches’ in which the primary goal was to strategize a potential vaccine design against Ebola virus variants using in-silico approaches. We identified a glycoprotein sequence which was similar in two Ebola variants and its antigenicity was checked using VaxiJen [hidden] which proved that the target sequence was antigenic in nature. Physicochemical and Structural analysis was performed and it was concluded that the selected target protein can be used as a best possible way to develop into a vaccine and for further research. My dissertation project at BHABHA ATOMIC RESEARCH CENTRE was- ‘To Investigate The Effect Of Redox Modifications On Functions Of [hidden] Cells’. I had hands-on training in various Animal tissue culture techniques like passaging, subculturing, thawing, freezing and cryopreservation, Maintenance and have used various high-tech instruments like IncuCyte, Flow cytometer, GelDoc, HTS analyzer, Microplate reader, Lyophilizer and BSL and animal hood. I have performed various phagocytosis assays, efferocytosis assays, PI staining, CFSE staining, DCFDA staining, NO assay, Clonogenic assay, MTT assay, ELISA, AGE, PAGE, RT-PCR, Cell cycle analysis, Bradford assay, Imaging. I am well-versed in using different cell-lines and their handling and storing them by making cryo vials. Apart from this I also had training in animal handling and mice dissection of spleen, intestine and bone marrow of BALB/c and Swiss mice. I had a working experience of 3 years in the Centre for cellular and molecular biology (CCMB) under the guidance of Dr. Puran Singh Sijwali and worked as a research associate in the Institute of Genomics and Integrative Biology (IGIB) under the guidance of Dr. Archana Singh for one year. I was working as Project scientist II in the Translational Health Science and Technological Institute (THSTI) under the guidance of Dr Milan Surjit.
    Research experience from [hidden] expertise (in research) belongs to the cloning and purification of proteins and their biophysical and computational approaches to investigate the molecular interactions of proteins to histone peptides. Most of my [hidden] research work focused on the UHRF1 (Ubiquitin-like, containing PHD and RING finger domain). We have demonstrated that the binding affinities of UHRF1 TTD exhibit higher binding affinity towardsH3K9me2 compared to H3K9me1/3through isothermal titration calorimetry (ITC). We have employed a computational tool, Molecular Dynamic (MD) simulation, to characterize the methyl status-specific binding of H3R2K9me by the TTD-PHD, and to analyze the effects of H3K4 di-methylation on H3R2K9me recognition. We have validated the MD simulation data through ITC binding study. These studies establish that the UHRF1 TTD-PHD exhibits lower binding free energy (ΔGbind) towards H3R2K9me2 compared to H3R2K9me3. We also showed that the SRA domain of UHRF1 binds to all 5mC oxidation derivatives.
    Research Associate: In CCMB, I was working on biochemical and biophysical characterization of DNA Damage inducible (DDi1) protein from Plasmodium falciparum. Ddi1 is a retroviral aspartyl protease and is involved in the repair of DNA-protein crosslink. Ddi1 also acts as a proteasome shuttle factor. We investigated the Plasmodium DDI1 to identify its roles during parasite development and potential as a therapeutic target. We have also knocked in pfDDi 1 into Saccharomyces cerevisiae to investigate the growth effect.
    In IGIB, I was working on isolation of Keratinocytes, Melanocytes and Fibroblast cells from foreskin patient samples and Cloning, Expression and purification of Transglutaminase 1(TGM1) protein from [hidden]
    In THSTI, I was involved in the Development of a recombinant vaccine against Hepatitis E Virus and their immunological characterization.
    Skills in Brief: I have also trained in molecular biology, biochemistry, and structural biology related techniques and also experienced in mammalian cell culture, Saccharomyces cerevisiae and Pichia pastoris.
