My college thesis project was titled- ‘Vaccine Design Against Ebola Virus Glycoprotein Using In-silico Approaches’ in which the primary goal was to strategize a potential vaccine design against Ebola virus variants using in-silico approaches. We identified a glycoprotein sequence which was similar in two Ebola variants and its antigenicity was checked using VaxiJen [hidden] which proved that the target sequence was antigenic in nature. Physicochemical and Structural analysis was performed and it was concluded that the selected target protein can be used as a best possible way to develop into a vaccine and for further research. My dissertation project at BHABHA ATOMIC RESEARCH CENTRE was- ‘To Investigate The Effect Of Redox Modifications On Functions Of [hidden] Cells’. I had hands-on training in various Animal tissue culture techniques like passaging, subculturing, thawing, freezing and cryopreservation, Maintenance and have used various high-tech instruments like IncuCyte, Flow cytometer, GelDoc, HTS analyzer, Microplate reader, Lyophilizer and BSL and animal hood. I have performed various phagocytosis assays, efferocytosis assays, PI staining, CFSE staining, DCFDA staining, NO assay, Clonogenic assay, MTT assay, ELISA, AGE, PAGE, RT-PCR, Cell cycle analysis, Bradford assay, Imaging. I am well-versed in using different cell-lines and their handling and storing them by making cryo vials. Apart from this I also had training in animal handling and mice dissection of spleen, intestine and bone marrow of BALB/c and Swiss mice. My project included 2 antioxidants (Chlorophyllin and Baicalein) and 2 prooxidants (Plumbagin and Mitocurcumin) and their modulations on the different functions of RAW cells [hidden] macrophages and how this could be applied further if there is a malfunctioning of these cells. I had my internship training in Apollo Diagnostics and had hands on training in RT-PCR techniques for covid-19 and Manual and automated RNA isolation techniques and data entry operations. I am currently working as a Project Assistant on an ICMR funded project- “Immunometabolomics in the quest for comprehending protection and pathogenesis in tuberculosis”. My work involves understanding metabolite profiles from patient immune cells, comparing them with healthy individuals and those with latent TB. In this role, I have gained hands-on experience in patient blood collection, Density gradient centrifugation, PBMC’s isolation and culture maintenance, Metabolite extraction, analyzing mass spectrum results, Isolation of T-cells by Magnetic associated cell separation, T-cell expansion, Intracellular staining for Flow cytometry, as well as REMA assay, Heat induced Hemolysis and Docking.
