    – Isolation of _MYB_ genes from high pungent _Capsicum chinense_ for cloning and overexpression, in low pungent _Capsicum annuum_ via Agrobacterium-mediated plant transformation, tissue culture (shoot and root regeneration), transgenic development and screening.
    – GFP tagging of MYB transcription factor proteins, agroinfiltration in _Capsicum_ leaf epidermis and subcellular nuclear localisation using confocal microscopy.
    – Interspecific crossing of contrasting pungency containing chilli pepper species (_Capsicum annuum _and_ Capsicum chinense_) and development of F2 population
    – Phenotyping- Extraction and estimation of capsaicinoids- capsaicin and dihydrocapsaicin in a large chilli pepper germplasm and F2 population using HPLC.
    – Genotyping- Identification, development and validation of molecular markers (SSRs, InDels and SNPs) in MYB gene and promoter sequences for marker-assisted selection of high pungency trait in large germplasm of chilli pepper varieties and F2 population.
    – Association analysis between phenotypic data and marker allele data using TASSEL.
    Beyond my doctoral work, I have actively engaged in collaborative research projects, such as:
    – Isolation of pungency biosynthesis pathway genes- _Amt, Kas_ and _Pun_ genes from highly pungent _Capsicum chinense_ and transgene expression in cherry tomato using GATEWAY cloning.
    – Whole proteome analysis of fruit developmental stages of high and low pungent _Capsicum_ species using LCMS-MS to identify differently abundant proteins.
    – Biolistics transformation of onion peel for protein-protein interaction studies of MYB transcription factors with pungency biosynthesis pathway proteins- Pun, Amt and Kas using BiFC.
    – Metabolite analysis of eggplant under nematode _(Meloidogyne incognita) _stress and alleviation (biocontrol _Ganoderma lucidium_ macrofungi) conditions using GCMS.
    – Metabolite analyses of _Capsicum_- roots, leaves, stem, and fruits under mineral (Phosphate, Iron and Zinc) element starvation and abundance stress via GCMS.
    – Extraction and estimation of terpenoids (Phytosterols) in _Premna herbaceae _leaves and roots_ _via GCMS.
    – Extraction and estimation of carotenoid and anthocyanins pigments in pepper fruits using HPLC.
    My recent postdoctoral research focused on integrating proteomics, transcriptomics and metabolomics to identify differentially abundant transcription factors involved in capsaicinoids biosynthesis in _Capsicum chinense_. This involved several methodologies such as:
    – Whole metabolite extraction from two fruit developmental stages of high and low pungent _Capsicum_ species and estimation and analysis via GCMS.
    – Correlation analysis between proteome, transcriptome and metabolome data to understand the complex capsaicinoid biosynthesis pathway in _Capsicum chinense_.
    Further to advance in my professional career, I am eager to join the Research Data Analyst position at IIT BHU, as it aligns with my research data analysis experiences.
