Worked on determining the efficacy of AAV dual vector editing in CD34+ cells in vivo. To demonstrate the best AAV serotype for CD34+ cell transduction in vivo and test different correction modes for sickle cell disease. Furthermore, to select the finest combination to go for clinical trials.

Doctoral Dissertation

My doctoral research focused on exploring the therapeutic potential of 3,3-diindolylmethane (DIM), a small molecule, as a novel inhibitor of the IL-17A/IL-17RA interaction for treating psoriasis. Unlike current IL-17A inhibitors, DIM aimed to target IL-17A signaling effectively, showcasing its promise as a multifaceted therapeutic agent for psoriasis.

D-NOME Biosciences
Hyderabad, Telangana, India
Dear Hiring Manager,
I am writing to express my interest in the position of **Scientist – Molecular Biology (B)** at D-NOME Biosciences. With a strong academic foundation (Ph.D. in Biological Sciences – Immunology from VIT), extensive laboratory experience, and postdoctoral work in gene therapy, I am confident in my ability to make meaningful contributions to your team.
My recent postdoctoral work at Narayana Nethralaya Foundation focused on AAV-mediated gene editing in CD34+ cells to develop curative strategies for sickle cell disease. This involved designing viral vectors, optimizing transduction protocols, and testing efficacy in in vivo models. During my Ph.D., I led a project exploring small-molecule inhibitors targeting IL-17A/IL-17RA interactions for psoriasis therapy, employing molecular cloning, protein expression, immunological assays, and flow cytometry. This dual exposure to both immunology and gene therapy has provided me with a unique, multidisciplinary perspective.
Key technical competencies that align with your requirements include:
· **Molecular cloning**, gene expression studies, and protein purification workflows.
· Experience with **flow cytometry**, Western blotting, qRT-PCR, ELISA, and immunohistochemistry.
· Skilled in **AKTA-based chromatography** and downstream analysis.
· Certified in **Biosafety Level 2**, genetic counseling, and animal handling (ICMR – NIN).
With a publication record in reputed journals and experience mentoring junior researchers, I am both a capable bench scientist and a collaborative team member. I am highly motivated to contribute to D-NOME’s mission of advancing molecular biology research and product development.
I have attached my CV for your consideration. I would be delighted to discuss how my skills and experiences can align with D-NOME’s goals. Thank you for your time and consideration.
Sincerely,
**Dr. Srikanth Manupati**
